Glossary
Terms used in planning, library QC, and abundance analysis.
- sgRNA
- Single-guide RNA used to target a gene in CRISPR-KO, CRISPRi, or CRISPRa screens.
- pegRNA
- Prime-editing guide RNA. Different pegRNAs can share a spacer but differ in RTT or PBS, so spacer alone is not a unique identity.
- Variant
- A defined genomic REF-to-ALT change. Prime Editing Build requires GRCh38 coordinates and checks the REF allele.
- MOI
- Mean effective viral integrations per cell in the Poisson planning model. At MOI 0.3, approximately 85.7% of infected cells are expected to carry one integration.
- Representation
- Number of cells, independent DNA templates, or sequencing reads per library reagent. Always check the unit and experimental stage.
- Biological replicate
- Independently conducted infection, culture, treatment, and harvest. Resequencing the same sample is a technical replicate.
- T0
- Baseline sample collected before the experimental endpoint comparison.
- gDNA
- Genomic DNA extracted from cells, containing integrated guide cassettes for abundance sequencing.
- Gini coefficient
- Summary of count inequality across a plasmid library; zero denotes perfectly equal counts.
- Log2 fold change
- Log2 ratio of normalized endpoint and baseline abundance for a reagent.
- FDR
- False discovery rate reported for a specified statistical analysis and contrast. It does not by itself validate a biological mechanism.
- PRIDICT2
- External pegRNA candidate-prediction engine; its score is not a measured editing fraction.
- pegLIT
- External engine for epegRNA linker engineering.
- MAGeCK
- External statistical software used for screen abundance analysis.
See Documentation for planning assumptions and References for external software and library sources.
