Transduction & MOI

At MOI 0.3, the Poisson model estimates 85.7% of infected cells carry exactly one sgRNA.
Minimum cellular coverage to maintain during culture. This is distinct from independent molecular templates recovered after gDNA extraction.

Measure this in a pilot kill curve for your exact cell line and vector.
Puromycin is commonly practical because killing is rapid; establish its dose in an untransduced-cell kill curve. The vector resistance or fluorescence marker must differ from any pre-existing cellular marker.
